Apoptose

Apoptose ist ein hochregulierter Prozess, mithilfe dessen sich Organismen auf organisiertem Weg nicht mehr benötigter Zellen entledigen können. Sie wird entweder durch eine bestimmte Klasse von Todesrezeptoren oder durch intrinsische Faktoren wie DNA-Schäden ausgelöst. Die Apoptose wird hauptsächlich von Caspasen angetrieben und weiterhin durch Bcl-2 und Inhibitorproteine reguliert. Eine Dysregulation dieses programmierten Zelltods wird oft mit Krebs in Verbindung gebracht.

Autophagie

Als Autophagie oder auch Autophagozytose wird gezielte der Abbau und die Verwertung von zelleigenen Bestandteilen bezeichnet. Hierbei werden sowohl kleine fehlgefaltete Proteine als auch ganze Organellen verwertet. Der lysosomale Signalweg initiiert das Umschließen, Abbau und Recycling der zellulären Komponenten, inklusive alternder Proteine und Organellen, und fördert so das Überleben der Zelle.

BCR Signaling

Der B-Zell-Rezeptor (BCR)-Signalweg ist essentiell für das Überleben und die Entwicklung von B-Zellen und die Antikörperproduktion unter physiologischen und pathologischen Bedingungen. Antigen-getriebene Priming-Signalisierung ist wichtig für die Initiierung der B-Zell-Aktivierung und Differenzierung in Antikörper-sezernierende Zellen. Auf der anderen Seite ist die tonische BCR-Signalisierung für das Überleben und die Entwicklung von B-Zellen erforderlich, während die chronische Signalisierung für die Proliferation von B-Zell-Lymphomzellen essentiell ist.

Cancer Immune Checkpoints

Immun-Checkpoints sind Rezeptoren auf der Membran von T-Lymphozyten, die deren Immunantwort abwechselnd steuern können. Sie regulieren somit die Immunantwort entweder über Stimulation oder Inhibition. Der Signalweg ist entscheidend, um die Selbsttoleranz zu fördern und Autoimmunität und damit verbundene Krankheiten zu verhindern.

DNA Reparatur

In jedem Organismus gewährleisten zahlreiche DNA-Reparaturmechanismen die Integrität der im Kern einer jeden Zelle enthaltenen genetischen Information. Die zentrale Bedeutung der DNA-Integrität wird anhand der zahlreichen unterschiedlichen Signalwege für die DNA-Reparatur sowie den verschiedenen an diesen Prozessen beteiligten Proteinen deutlich.

Inflammasome

Inflammasome sind Multiproteinkomplexe, die sich im Zytosol befinden. Typischerweise bestehen sie aus einem Sensorprotein, einem Adaptorprotein, das eine Caspase-Rekrutierungsdomäne enthält, und einer pro-inflammatorischen Caspase. Im Falle des NLRP3-Inflammasoms, dem derzeit am besten charakterisierten Inflammasom, sind die entsprechenden Proteine NLRP3, ASC/PYCARD und CASP1.

T-Zell Rezeptor Signalweg

TLR Signalweg

Toll-like-Rezeptoren spielen eine wesentliche Rolle bei der angeborenen Immunantwort gegen verschiedene Pathogene wie Viren, Bakterien und Pilze. TLRs lösen einen von zwei konservierten Entzündungspfaden aus: entweder den TRIF- oder den MyD88-abhängigen Pfad. Außerdem induziert die TLR-Signalisierung auch die Reifung dendritischer Zellen und die adaptive Immunantwort.

Ubiquitin Proteasome Pathway

Der Ubiquitin-Proteasom-Weg ist für den gezielten Abbau der meisten kurzlebigen Proteine in der eukaryontischen Zelle erforderlich. Im Gegensatz zu den meisten Regulationsmechanismen ist der Proteinabbau von Natur aus irreversibel. Die Zerstörung eines Proteins kann zu einer vollständigen, schnellen und dauerhaften Beendigung des Prozesses führen, an dem das Protein beteiligt ist, sowie zu einer Veränderung der Zellzusammensetzung. Der schnelle Abbau bestimmter Proteine ermöglicht die Anpassung an neue physiologische Bedingungen.

Die Unfolded Protein Response (UPR) ist eine komplexe Stressreaktion von Zellen, ausgelöst durch die Ansammlung von Proteinen mit fehlerhafter Faltung im endoplasmatischen Retikulum (ER).
