Hintergrund
HIPPI was identified as a protein that interacts with HIP1 (Huntingtin-interacting protein 1). HIPPI mRNA is expressed in the brain. It is present in all regions of the brain, and to a lesser extent in spinal cord. It is also expressed abundantly in heart, kidney, and lung, with weaker expression in all other tissues tested. HIPPI is expressed in the Golgi apparatus of neurons where it partly colocalizes with HIP1. HIPPI forms a heterodimer with HIP1. The availability of free HIP1 is modulated by polyglutamine length within huntingtin, with disease-associated polyglutamine expansion favoring the formation of proapoptotic HIPPI-HIP1 heterodimers. These heterodimers can recruit procaspase-8 into a complex of HIPPI, HIP1, and procaspase-8 and launch apoptosis through components of the extrinsic cell death pathway.
