ASAP (Arf-GAP with SH3 domain, ANK repeat and PH domain-containing protein 1) or Development and differentiation-enhancing factor 1 or DDEF1 is an ADP-ribosylation factor (ARF) GTPase-activating protein with two known isoforms: ASAP1a and ASAP1b (125 and 126 kDa, respectively). ASAP contains a pleckstrin homology (PH) domain, a zinc finger motif, 3 ankyrin repeats, a proline-rich region containing several SH3 ligand motifs, and an SH3 domain. GTPase-activating property of ASAP1 is induced by phosphatidylinositol 4,5-biphosphate (PIP2) of the PH domain. ASAP1 plays an important role in cytoskeleton remodeling and regulation of membrane trafficking. ASAP1 acts as a proto-oncogene in breast cancer and other malignant tumors. ASAP is highly expressed in brain tissue.
