This mouse TNFalpha Enzyme Immunometric Assay (EIA) kit is a complete kit for the determintation of mouse TNFalpha in biological fluids. Please read the complete kit insert before performing this assay. The kit uses a monoclonal antibody to mouse TNFalpha immobilized on a microtiter plate to bind the mouse TNFalpha in the standards or sample. A recombinant mouse TNFalpha Standard is provided in the kit. After a short incubation the excess sample or standard is washed out and a polyclonal antibody to mouse TNFalpha is added. This antibody binds to the mouse TNFalpha captured on the plate. After a short incubation the excess antibody is washed out and donkey antirabbitIgG conjugated to Horseradish peroxidase is added, which binds to the polyclonal mouse TNFalpha antibody. Excess conjugate iswashed out and substrate is added.After a short incubation, the enzyme reaction is stopped and the color generated is read at 450 nm. The measured optical density is directly proportional to the concentration of mouse TNFalpha in either standards or samples.

Tumor Necrosis Factoralpha (TNF alpha) is a 17.5 kDa, 157 amino acid protein that is a potent lymphoid factor, which exerts cytotoxic effects on a wide range of tumor cells and other target cells. TNF alpha has been suggested to play a proinflammatory role and has been detected in synovial fluid of patients with rheumatoid arthritis. It is the primary mediator of immune regulation. The biosynthesis of TNF alpha is tightly controlled, being produced in extremely small quantities in quiescent cells, but is a major secreted factor in activated cells.
