Mouse pre-B-cell line 300. 19, stably transfected with the full length Human Ly-9 cDNA. Remarks: Hybridoma was established by fusion of mouse myeloma cell NS-1 with Balb/cmouse splenocyte.
Flow Cytometry: 10 μg/mL (final concentration). Positive Control: Peripheral blood T cells. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.

Flow Cytometric Analysis for Floating CellsProtocol 1We use Fisher tubes or equivalents as reaction tubes for all step described below. 1) Wash the cells 3 times with washing buffer [PBS containing 2% fetal calf serum (FCS) and0. 1% BaN3]2) Add 10 µL of normal goat serum to the cell pellet after tapping. Mix well, and incubatefor 5 minutes at room temperature (20~25°C). 3) Add 30 µL of the FITC labeled CD229 monoclonal antibody (10-20 µg/mL) diluted withPBS containing 2% FCS. Mix well, and incubate for 30 minutes at room temperature(20~25°C). 4) Add 1 mL of the washing buffer followed by centrifugation at 500xg for 1 minute at RT(20~25°C). Remove supernatant by careful aspiration. 5) Resuspend the cells with 500 µL of the washing buffer and analyze by a flow cytomete.

CD229 is a 100-120 kD cell surface glycoprotein that is the homologue of murine Ly-9. CD229 is a member of the immunoglobulin superfamily and may have a function in cell signalling. The antigen is expressed at low levels by mature T and B lymphocytes and at higher levels by single-positive thymocytes.
