Hintergrund
- IL6 exerts its activity through binding to a high affinity receptor complex consisting of two membrane glycoproteins: an 80 kDa IL6 binding receptor protein (IL6R) and a 130 kDa signal transducing protein consisting of a 19 amino acid residue signal peptide (gp130). Human IL6R is a 468 amino acid residue type I membrane protein consisting of a 19 amino acid residue signal peptide, a 339 residue extracellular region, a 28 residue transmembrane region and an 82 residue cytoplasmic region. The ectodomain of IL6R is made up of an immunoglobulin like domain and a cytokine receptor superfamily domain. A naturally occurring soluble form of the IL6R, which binds IL6 and which mediates IL6 signaling through interaction with gp130, is identified in the urine of healthy adult humans. It has also been detected in human and mouse serum as well as in the conditioned medium of various cells including human peripheral blood mononuclear cells, and several T cell or granulocyte/macrophage cell lines.
