Usage: Western Blot: 0.5-2 mg/mL detects STAT5B (doublet of 90kD and 92kD) in. human A431 and mouse 3T3 fibroblasts using enhanced chemiluminescence with a five minute exposure. Immunoprecipitation: 4 μg immunoprecipitates STAT5B from A431 carcinoma cells and a. previous lot immunoprecipitated STAT5B from human Raji cells. A RIPA lysate of human A431 cells (500 μg protein) was incubated with 4 μg of anti-STAT 5B Immune complexes were precipitated with protein A agarose and analyzed by immunoblotting with anti- STAT 5B. Bands were visualized using a goat anti-rabbit IgG secondary antibody conjugated to HRP in conjunction with enhanced chemiluminescence detection. Positive Antigen Control: mouse 3T3 cell lysate. Add 2.5 μL of 2-mercaptoethanol. per 100 μL of lysate and boil for 5 minutes to reduce the preparation. Load 20 μg of reduced lysate per lane for minigels.
