Anwendungsinformationen

Applikationshinweise: Neutralisation: To yield one-half maximal inhibition [ND50] of the biological activity of hPDGF-AA (5.0 ng/mL),a concentration of 0.05 - 0.07 μg/mL of this antibody is required. Immunohistochemistry: The antibody stained formalin-fixed, paraffin embedded sections of normal human skin at1.0 μg/mL overnight at 4C. (A HRP-labeled polymer detection system was used with anon-alcohol soluble AEC chromogen and a proteinase K antigen retrieval). ELISA: Indirect - To detect hPDGF-AA by indirect ELISA (using 100 μL/well antibody solution),a concentration of 0.5 - 2.0 μg/mL of this antibody is required, allows the detection of atleast 0.2 - 0.4 ng/well of recombinant hPDGF-AA. Sandwich - To detect hPDGF-AA by sandwich ELISA (using 100 μL/well antibody solution),a concentration of 0.5 - 2.0 μg/mL of this antibody is required, in conjunction with abiotinylated anti-human PDGF-AA (PP1061B) as the detection antibody at least 0.2 - 0.4 ng/well of recombinant hPDGF-AA can be detected. Western blot: To detect hPDGF-AA by Western blot analysis this antibody can be used at a concentrationof 0.1 - 0.2 μg/mL. Used in conjunction with compatible secondary reagents the detectionlimit for recombinant hPDGF-AA is 1.5 -3.0 ng/lane, under either reducing or non-reducingconditions.
Other applications not tested.
Optimal dilutions are dependent on conditions and should be determined by the user.
