Notch Signalweg

Der hoch konservierte Notch-Signalweg ist einzigartig, da es sich beim Notch-Rezeptor und einem Großteil seiner Liganden (Mitglieder der DSL(Delta/Serrate/Lag-2)-Familie) um Transmembranproteine handelt, die an der Zelloberfläche gebunden sind. Dadurch ist die Notch-Signalübertragung auf die Interaktion zwischen angrenzenden Zellen begrenzt. Die Kommunikation zwischen benachbarten Zellen ist von größter Bedeutung, insbesondere während der frühen Zell-Entwicklung, wenn die Funktion der Zelle noch nicht bestimmt ist. Der Notch-Signalweg ermöglicht Zellen, ihre eigene Identität festzulegen und gleichzeitig die Funktion und Identität von Nachbarzellen durch laterale Hemmung zu beeinflussen.

Der Kern des Notch-Signalwegs umfasst zwei angrenzende Zellen, wovon die eine einen Liganden der DSL-Familie exprimiert und die andere den Notch-Rezeptor. Wenn der Rezeptor und der Ligand sich verbinden, spalten zwei getrennte Proteasen den Notch-Rezeptor in extrazelluläre und zytosolische Komponenten. ADAM10 spaltet den extrazellulären Teil des Notch-Rezeptors, welches an seinen zugehörigen Liganden gebunden bleibt und von der Liganden-exprimierenden Zelle endozytiert wird. Gamma-Sekretase spaltet den zytosolischen Teil des Notch-Rezeptors. Dieser wandert zum Zellkern, wo er an den Transkriptionsfaktor CSL bindet. Dieser wiederum wandelt ihn von einem transkriptionellen Repressor in einen Aktivator und reguliert die Exprimierung von Notch-Zielgenen hoch.
