Beschreibung	CD14 is a 53'000 - 55'000 Mr glycoprotein strongly expressed on the surface of monocytesand weakly on the surface of granulocytes. It is anchored to the surface by linkage toglycosyl phosphatidylinositol (GPI). CD14 functions as a receptor for endotoxin and is themajor receptor involved in the lethal response to both endotoxin and Gram-negativebacteria. In addition to membrane CD14, soluble forms of CD14 can be detected in serumand tissue culture supernatants of cells transfected with CD14.
Spezifität	Monoclonal antibody X-8 prevents the Lipopolysaccharide Binding Protein (LBP)independent activation of cells via soluble CD14 by blocking the binding of CD14.

Applikationshinweise	Suitable for Immunohistochemistry (IHC) on frozen sections (0.25-0.5 ug/ml,1/800-1/1600) and FACS. Suggested positive control: Human placenta. Antigen Distribution on Isolated cells and Tissue sections: Monocytes, granulocytes, macrophages in myeloid leukaemia. Cell Lines: HL-60 without stimulation negativeHL-60 stimulated with PMA 15% positiveHL-60 stimulated with DMSO 15% positiveU937 negativeX-8 is positive on Langerhans cells, follicular dendritic cells, histiocytes and highendothelial cell venules. Functional assays. Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
