    Hintergrund
    Parkin is a zinc-finger protein with an amino-terminal ubiquitin domain and a carboxy-terminal RING-box. The encoded protein is a component of a multiprotein E3 ubiquitin ligase complex that mediates the targeting of substrate proteins for proteasomal degradation. Phosphorylation of Parkin at Ser101 has a regulatory role on its E3 ubiquitin ligase activity. Mutations in this gene cause Parkinson disease and autosomal recessive juvenile Parkinson (AR-JP) disease which is characterized by the progressive loss of dopaminergic neurons and the presence of Lewy bodies in the substania nigra. Alternative splicing of this gene produces multiple transcript variants encoding distinct isoforms. Parkin is an ubiquitin-protein ligase expresses in many tissues such as brain, testis, skeletal muscle and heart. In humans, Parkin is highly expressed in the basal ganglia, midbrain, cerebellum and cerebral cortex. Parkin expression is also high in the brainstem of mice, with the majority of immuno positive cells being neurons. It is a RING-type E3 ubiquitin-protein ligase that facilitates the ubiquitination of misfolded proteins by participating in the removal and/or detoxification of abnormally folded or damaged protein. It also plays a role in controlling neurotransmitter trafficking at the presynaptic terminal and in calcium-dependent exocytosis. Parkin is an E3 ubiquitin ligase used to reduce intra neuronal amyloid-β (Aβ) levels and enhance autophagic clearance of Aβ-induced defects in AD. Parkin-mediated clearance of ubiquitinated Aβ act in parallel with autophagy to clear molecular debris and defective mitochondria and restore neurotransmitter balance.The human Parkin gene maps to chromosome 6q25.2-q27 and encodes a 465 amino acid protein.
