Applikationshinweise: Before using Kit, spin tubes and bring down all components to bottom of tube. Duplicate well assay was recommended for both standard and sample testing.

Protokoll: ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. An antibody has been precoated onto 96-well plates. Standards and test samples are added to the wells, a biotinylated detection antibody specific for target is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the target amount in sample captured in plate.

Background: Insulin-like growth factor-binding protein 6 (IGFBP6) is a protein that in humans is encoded by the IGFBP6 gene. The IGFBP6 gene is mapped to chromosome 12. In all, it has contained 216 amino acid protein with a calculated molecular mass of 22,847, The human keratinocyte cell line HaCat secretes IGFBP6 as an autocrine growth inhibitor. Recombinant IGFBP6 was also shown to inhibit growth of HaCat cells and other keratinocyte cell lines.
