  • Applikationshinweise
    This affinity purified antibody detects human PDK-1 and has been tested for use in ELISAand immunoblotting. Although untested, this reagent may be useful for Immunohistochemistry andImmunoprecipitation. It recognizes 60,000 MW human PDK-1. Reactivity in otherimmunoassays is unknown. Recommended Dilutions: This product has been assayed by immunoblot using HRPGoat-anti-Rabbit IgG [H&L] and Enhanced Chemiluminescence for detection. A workingdilution range of 1: 750 is suggested to detect PDK-1 in lysates derived from Sf9 insect cellsinfected with virus expressing myc-tagged human PDK1. In this experiment, approx. 10 μg of bothan untransfected sf9 cell lysate (not shown) and PDK-1 virus-infected Sf9 lysate wereseparated on a 10 % gel by SDS-PAGE followed by transfer to nitrocellulose. Primaryantibody reaction proceeded overnight at 4C using a 1: 750 dilution of the antibody in 1 %non-fat milk. This antibody predominantly recognizes a 60 kDa band corresponding tohuman PDK-1.

    Hintergrund
    PDK-1 (3-Phosphoinositide-Dependant Protein Kinase-1) phosphorylates AGC kinases. PDK-1 activates conventional PKC and PKCz (zeta) through phosphorylation of critical threonine residues in the activation loop. PDK-1 also phosphorylates Protein Kinase B (PKB) at threonine 308 in the presence of phosphatidylinositol-3,4,5-trisphosphate. Active Akt inactivates Glycogen Synthase Kinase-3 (GSK3), eventually leading to the dephosphorylation and activation of glycogen synthase, and the stimulation of glycogen synthesis.
