Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of mIL-3 (0.5 ng/mL), aconcentration of 0.15-0.20 μg/mL of this antibody is required. ELISA: (Indirect): To detect mIL-3 by indirect ELISA (using 100 μL/well antibody solution) aconcentration of 0.5-2.0 μg/mL of this antibody is required. This antigen affinity purifiedantibody, in conjunction with compatible secondary reagents, allows the detection of atleast 0.2-0.4 ng/well of recombinant mIL-3. (Sandwich): To detect mIL-3 by sandwich ELISA (using 100 μL/well antibody solution) aconcentration of 0.5-2.0 μg/mL of this antibody is required. This antigen affinity purifiedantibody, in conjunction with Biotinylated Anti-Murine IL-3 (PP010B) as a detectionantibody, allows the detection of at least 0.2-0.4 ng/well of recombinant mIL-3. Western Blot: To detect mIL-3 by Western Blot analysis this antibody can be used at a concentration of0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagents the detection limitfor recombinant mIL-3 is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

IL3 (Interleukin 3) is a 15 kDa cytokine that is primarily secreted by activated T lymphocytes and stimulates the proliferation and differentiation of hematopoietic cells. IL3 acts in hematopoiesis by controlling the production, differentiation, and function of two related white cell populations of the blood, the granulocytes and the monocytes/macrophages. It induces granulocytes, macrophages, mast cells, stem cells, erythroid cells, eosinophils and megakaryocytes and is expressed by activated T cells, mast cells, and natural killer cells. IL3 not only supports growth of both pluripotent stem cells and the more differentiated committed progenitors, but it also stimulates the functional activity of some fully differentiated cells.
