Rekonstitution
- Centrifuge the vial at 10,000 x g for 5 minutes before opening to ensure all material is collected at the bottom. The lyophilized product may be difficult to visualize. Add solvent directly to the centrifuged vial and dissolve by gently tapping, tilting, or rolling the vial. Avoid vigorous vortexing, a brief, light vortex of 1-2 seconds is acceptable if necessary. The product is soluble in DMSO. Prepare a concentrated stock solution by dissolving the lyophilized product in high-quality DMSO first (e.g., 100x to 1000x the final working concentration). Once fully dissolved, slowly dilute the stock solution into the desired aqueous buffer (or water) to the final working concentration while gently agitating the mixture. Centrifuge all product preparations before use. It is recommended to prepare fresh working solutions just before use. Avoid multiple freeze-thaw cycles to maintain optimal biological activity. Keep the DMSO concentration as low as possible. For cell-based assays, a final concentration of 0.1 % -0.5 % DMSO (v/v) is generally considered safe and non-toxic. For other experiments, a final concentration of 5 % DMSO (v/v) is usually acceptable. We advise checking relevant literature for acceptable concentrations for your specific cell line or assay system.
