    This ELISA allows quantification of albumin in mouse urine, so that an estimation of kidney function is possible.

    Determination of albumin is carried out as direct competitive ELISA.

    Protokoll

    Mouse albumin has been pre-coated onto a microtiter plate. During incubation binding of an enzyme-linked anti-albumin(mouse)-antibody is inhibited by albumin from samples/ standards. After washing away any unbound antibody a substrate solution is added. Developing colour correlated with the amount of antibody conjugate. Absorption at 450 nm is reverse proportional to albumin concentration.

    Testdurchführung

    Incubation of samples/standards with antibody conjugate: 50 µL + 50 µL/well 90 min with shaking,120 min without shaking.
    Wash Substrate incubation: 100 µL/well 10 min, room temperature.
    Add stop solution 100 µL/well.
    Read at 450 nm

    Testpräzision

    Intraassay precision (CV):(sample dilution 1:100, n=12) at 3.1 µg/ml: 14.3%at 30.4 µg/ml: 6.1% at 223 µg/ml: 5.8%

    When blood is filtered in glomeruli of kidneys, highly molecular substances are held back. Therefore, urine is nearly protein free. In case of kidney damages, small proteins like albumin appear in urine. Albumin therefore serves as a marker in monitoring kidney damages. In examining nephrotoxic substances and therapeutic strategies regarding kidney damages, animal models are frequently used.
