    Spezifität

    This antibody recognizes and binds the beta chain of the IL-2 receptor and completely inhibits the IL-2 binding to the beta chain. The beta subunit is involved in receptor mediated endocytosis and transduction of the mitogenic signals of IL2.

    The original antibody was generated by immunizing BLAB/c mice with YTS cells, a human NK-like cell line that highly expresses the human IL-2R beta chain as immunogen.

  • Applikationshinweise

    Mik-beta 1 inhibited binding of iodine labelled IL-2 to YTS cells in a dose dependent manner. Mik-beta 1 also successfully immunoprecipitated beta chain of the IL-2R from radio iodinated YTS extract. Biotinylated Mik-beta 1 also efficiently competes with IL-2 for binding to solubilized beta chain. Flow cytometry was used to study the cellular disctribution of beta chain, as determined by mik-beta 1 staining. This antibody completely inhibited IL-2 dependent T cell growth in presence of anti-Tac. Two-color flow cytometric analysis using anti-beta chain mAbs clearly showed that approximately 10 % of peripheral blood lymphocytes manifest the beta chain and that this population is mainly composed of Leu-19+ NK cells (PMID: 2467293). Mik beta 1 antibody acts synergistically with humanized anti-Tac to prevent IL-2-induced proliferation of activated T cells (PMID: 8335891). A recombinant single chain immunotoxin of Mik-beta 1 and Pseudomonas derived exotoxin PE40 was selectively cytotoxic for cells expressing high levels of IL-2R beta (p75) subunit (PMID: 1401913).

    This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG2a format, for improved compatibility with existing reagents, assays and techniques.
