    Anti-Transferrin receptor [RI7 217.1.3], Rat IgG2a, kappa

    Spezifität

    The transferrin receptor is necessary for cellular iron uptake by the process of receptor-mediated endocytosis. Mice that are deficient in this receptor show impaired erythroid development and abnormal iron homeostasis.

    Application Notes original Clone: This antibody was generated and used in a flow cytometry assay to identify murine hematopoietic progenitor cells expressing the transferrin receptor (Lesley et al., 1984, PMID: 6319033). The original format of this antibody was tested and found to moderately reduce cell growth. A Fab version of this antibody was generated, its binding was tested, and it was compared to its full antibody counterpart. In contrast to the original antibody format, the Fab fragment did not cause receptor down-regulation or accelerated degradation. Both the Fab fragment and the full antibody were successfully used in an IF staining assay (Lesley et al., 1989, PMID: 2653853). The original fotmat of this antibody was demonstrated to undergo receptor-mediated transcytosis across the blood-brain barrier (BBB), as confirmed by capillary depletion studies, and to effectively transport into the mouse brain in vivo (Lee et al., 2000, PMID: 10688622). Injection of this antibody into mice resulted in CD71+ erythroid splenocyte cell depletion (Torow et al., 2015, PMID: 26195040).
