Applikationshinweise
- Detection of guinea pig IFN-γ producing cells by flow cytometry. 5 μL is recommended for staining of 1 million cells in a total volume of 50 μL. PF488P is excited by the blue laser (488 nm). The excitation max is 490 nm and the emission max is 516 nm. Detection of IFN-γ by flow cytometry in viable guinea pig spleen cells. Cells were stimulated for 16 hrs in the presence of PMA/ ionomycin and Brefeldin A. Cells were then fixed and permeabilized using 4 % paraformaldehyde and saponin, and subsequently stained with MT4A27-PF488P (solid line). Matched isotype control antibody (dashed line). The histogram derives from gated events of typical lymphocyte characteristics in forward and side light scatter. Flow cytometry was performed on a BD FACSVerse system.
