    Immunohistochemistry Dilution: 1:50

    Application Note: Anti-PhosphoTyrosine (pY) Antibody is tested for use in ELISA, western blotting, immunohistochemistry-P, immunoprecipitation, and Non-radioactive Tyrosine kinase assays.  Anti-PhosphoTyrosine (pY) Antibody reacts specifically with phosphotyrosine and shows minimal reactivity by ELISA and western blotting with proteins containing phosphothreonine or phosphoserine residues. Anti-PhosphoTyrosine (pY) Antibody reacts with free phosphotyrosine, phosphotyrosine conjugated to carriers such as thyroglobulin or BSA, and detects the presence of phosphotyrosine in proteins of both unstimulated and stimulated cell lysates.  Although not tested, Anti-PhosphoTyrosine (pY) Antibody is likely functional in RIA and flow cytometry.

    Store PhosphoTyrosine antibody at -20° C prior to opening. Aliquot contents and freeze at -20° C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use.

    Background: Phosphotyrosine is the tyrosine amino acid that has been phosphorylated and is considered one of the integral steps in signal transduction and general enzyme activity. Tyrosine is phosphorylated through protein kinases and through phosphorylation, phosphotyrosine can bind other proteins causing binding of multi-protein complexes that connect the upstream kinases and downstream effectors.
