1. Staining of formalin-fixed tissues requires boiling tissue sections in 10  mM Tris buffer with 1  mM EDTA,  pH 9.0, for 10-20 min followed by cooling at RT for 20 min.\. Flow Cytometry: 0.5-1 μg/million cells in 0.1ml,Immunofluorescence: 0.5-1 μg/mL,Western blot: 0.5-1 μg/mL,Immunohistochemistry (FFPE): 2-4 μg/mL for 30 min at RT (1)

Hintergrund: CD86 is a receptor involved in the costimulatory signal essential for T-lymphocyte proliferation and interleukin-2 production, by binding CD28 or CTLA-4. May play a critical role in the early events of T-cell activation and costimulation of naive T-cells, such as deciding between immunity and anergy that is made by T-cells within 24 hours after activation. Isoform 2 interferes with the formation of CD86 clusters, and thus acts as a negative regulator of T-cell activation. [UniProt]
