Beschreibung: ATF3, or ActivatingTranscription Factor 3, is a member of mammalianactivation TF/CREB protein family of transcriptionfactors. ATF3 binds the cAMP response element (cre)(consensus: 5'-gtgacgt[ac][ag]-3'), a sequence present inmany viral and cellular promoters. However, ATF3represses rather than activates transcription frompromoters with ATF sites stabilizing inhibitory co-factorsat the promoter. Alternate splicing forms of ATF3, calledATF3 delta Zip, lack the leucine zipper domain and donot bind DNA. ATF3 delta Zip stimulates transcription,presumably by sequestering inhibitory co-factors awayfrom the promoter. Human ATF3 (SwissProt 18847) is a20575 Da protein composed of 181 amino acids.

Spezifität: This affinity-purifiedantibody is directed against human ATF3 and isuseful in determining its presence in various assaysincluding ELISA and western blotting. Thispolyclonal antibody recognizes over-expressedATF3 protein found in various expression systems.Reactivity is observed against human ATF3. Crossreactivity with ATF3 from other mammalian sourceshas not been tested.

Anwendungen

Applikationshinweise: Recommended Dilutions: This product was assayed by immunoblot and was found to be reactive againstrecombinant ATF3 present in transfected mammalian or E.coli whole cell extracts. For western blot a 1:200dilution yields strong signal with relatively clean background staining. Use IRDYE800 Conjugated Affinity Purified Anti-RABBIT IgG (H&L) (GOAT) Min X Bv Ch Gt GP Hs Hu Ms Rt & Sh Serum Proteins (code 611-132-122) as a secondary antibody for detection at a 1:5,000 dilution. LICOR's Odyssey Infrared Imaging Systemwas used to scan and process the image. For ELISA a 1:5,000 to 1:20,000 dilution gave strong signal against theimmunizing peptide. Similar dilutions should generate sufficient ELISA signal against native or recombinant ATF3protein. The researcher should determine optimal titers for other applications. Note: Affinity purified anti-ATF3 has been tested by ELISA and western blotting againstrecombinant forms of the protein. Although not tested, this antibody is likely function in most immunoassaysincluding immunofluorescence microscopy, immunohistochemistry.
