Produktbeschreibung

Immunogen This affinity purified antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding to amino acids 301-312 of PDK-1 (genome data base accession number AC005591). Peptide sequence: R-K-I-Q-E-V-W-R-Q-R-Y-Q-S
Format Liquid (sterile filtered)
Beschreibung PDK-1 (3-Phosphoinositide-Dependant Protein Kinase-1) phosphorylates AGC kinases. PDK-1 activates conventional PKC and PKCz (zeta) through phosphorylation of critical threonine residues in the activation loop. PDK-1 also phosphorylates Protein Kinase B (PKB) at threonine 308 in the presence of phosphatidylinositol-3,4,5-trisphosphate. Active Akt inactivates Glycogen Synthase Kinase-3 (GSK3), eventually leading to the dephosphorylation and activation of glycogen synthase, and the stimulation of glycogen synthesis. Because of the role that PDK1 plays in insulin-induced glycogen synthesis and PKC activation, it is a potentially important target for metabolic drug research.
Spezifität This is an affinity purified antibody produced by immunoaffinity chromatography using the immunizing peptide after immobilization to a solid phase. This antibody is primarily directed against 60,000 MW human PDK-1 and is useful in determining its presence in various assays. Reactivity with PDK-1 from mouse or other sources has not been determined.

Anwendungen

Applikationshinweise Recommended Dilutions: This product has been assayed by immunoblot using HRP Goat-anti-Rabbit IgG [H&L] (code ABIN101990) and Enhanced Chemiluminescence for detection. A working dilution range of 1:750 is suggested to detect PDK-1 in lysates derived from Sf9 insect cells infected with virus expressing myc-tagged human PDK1. In this experiment, ~ 10 ug of both an untransfected sf9 cell lysate (not shown) and PDK-1 virus-infected Sf9 lysate were separated on a 10% gel by SDS-PAGE followed by transfer to nitrocellulose. Primary antibody reaction proceeded overnight at 4
Konzentration 1.3 mg/ml (by UV absorbance at 280 nm)
Buffer Stabilizer: None. Preservative: 0.01% (w/v) Sodium Azide. Buffer: 0.02 M Potassium Phosphate, 0.15 M Sodium Chloride, pH 7.2.
