Protein Background: PRDM16 (Histone-lysine N-methyltransferase PRDM16), also known as CMD1LL, KMT8F, LVNC8, MEL1 and PFM13. PRDM16 was found in t(1,3)(p36,q21)-positive AML, which has similar clinicopathological features to3q21q26 syndrome, as a transcriptionally activated gene near the chromosomal breakpoint. The structure of the PRDM16 gene product is very similar to that of MDS1/EVI1. Both products have the PRdomain at the amino-terminus, 2 separated DNA binding domains with 7 and 3 tandem zinc finger repeats, CtBP-binding motifs, and other possible transcriptional regulatory domains. By similarity, PRDM16 probably catalyzes the monomethylation of free histone H3 in the cytoplasm which is then transported to the nucleus and incorporated into nucleosomes where SUV39H methyltransferases use it as a substrate to catalyze histone H3 'Lys-9' trimethylation.
Short Description: Recombinant PRDM16 protein was expressed in a baculovirus expression system as the full length protein (accession number NP_071397.3) with an N-terminal FLAG tag. The molecular weight of the protein is 141.5 kDa.
