    Protein Background: The CDK1 / Cyclin B1 complex is a master regulator of the G2/M transition and progression through mitosis in eukaryotic cells. As a cyclin-dependent kinase complex, it phosphorylates numerous substrates to drive critical mitotic events, including nuclear envelope breakdown, chromosome condensation, spindle assembly, and activation of the anaphase-promoting complex/cyclosome (APC/C) for sister chromatid separation. Beyond its canonical cell cycle role, CDK1 / Cyclin B1 localizes to mitochondria and phosphorylates complex I proteins to enhance ATP production, thereby coupling energy generation with division demands. It also maintains chromosomal stability by regulating kinetochore microtubule attachments and spindle checkpoint signaling through phosphorylation of Mps1 and Aurora B kinases. In oocyte meiosis, the complex is essential for the metaphase I-to-anaphase I transition by promoting APC/C activation and homologous chromosome segregation. Dysregulation of CDK1 / Cyclin B1 contributes to cancer pathogenesis, as its overexpression or defective function (e.g., impaired Cyclin B1 induction) is linked to tumor proliferation, chemoresistance (e.g., T-DM1 in breast cancer), and genomic instability, making it a significant therapeutic target.

    Short Description: Recombinant CDK1 / Cyclin B1 Complex that includes full length human CDK1 protein (accession number NP_001777.1) with an C-terminal His tag and full length human Cyclin B1 protein (accession number NP_114172.1) with an C-terminal FLAG tag was expressed in Hi5 cells. The molecular weights of CDK1 and Cyclin B1 are 35.73 kDa and 50.03 kDa, respectively.
