    Testdurchführung

Adrenaline (epinephrine) and noradrenaline (norepinephrine) are extracted by using a cisdiolspecific affinity gel, acylated and then derivatized enzymatically. The competitive ELISA kit uses the microtiter plate format. The antigen is bound to the solid phase of the microtiter plate. The derivatized standards, controls and samples and the solid phase bound analytes compete for a fixed number of antiserum binding sites. After the system is in equilibrium, free antigenand free antigenantiserum complexes are removed by washing. The antibody bound to the solid phase is detected by an antirabbit IgGperoxidase conjugate using TMB as a substrate. The reaction is monitored at 450 nm. Determination of unknown samples is achieved by comparing their absorbance with a reference curve prepared with known standard concentrations.

    Hintergrund
Epinephrine (also known as adrenaline) is a hormone and a neurotransmitter. It increases heart rate, constricts blood vessels, dilates air passages and participates in the fight-or-flight response of the sympathetic nervous system. Chemically, epinephrine is a catecholamine, a monoamine produced only by the adrenal glands from the amino acids phenylalanine and tyrosine. Norepinephrine or noradrenaline is a catecholamine with multiple roles including as a hormone and a neurotransmitter. Areas of the body that produce or are affected by norepinephrine are described as noradrenergic. Norepinephrine is released from the adrenal medulla into the blood as a hormone, and is also a neurotransmitter in the central nervous system and sympathetic nervous system where it is released from noradrenergic neurons in the locus coeruleus.
