    Testdurchführung

    Canine sera for testing are diluted to 1:500 and allowed to react with pneumococcal Cpolysaccharide coated onspecially treated microwells. After appropriate incubation, the wells are washed to remove unreacted serum proteins,and an enzymelabeled goat antidog CRP (conjugate) is then added to react with and tag the antigenantibodycomplexes. Following another incubation period, the wells are again washed to remove unreacted conjugate. A ureaperoxide substrate with TMB as chromogen is added to start color development. Development of a blue color indicatesa positive reaction while negative reactions appear colorless or with a trace of blue. The reaction is interrupted with astop solution that turns the blue positive reactions to yellow. Negative reactions remain colorless or with a hint ofyellow. Color intensity (absorbance) is read at a wavelength of 450nm on a spectrophotometer or ELISA reader. Assigning a value for the absorbance can be accomplished by the use of a standard curve generated by measuring fourfold dilutions of the standard provided.

    Hintergrund

    Creactive protein (CRP) is a pentameric acute phase reactant that is synthesized by the liver. Its production is controlled primarily by interleukin 6. The serum CRP concentration may increase by up to 1000-fold with infection, trauma, surgery, and other acute inflammatory events. Chronic inflammatory disorders such as autoimmune diseases and malignancy can produce persistent high levels of serum CRP. Traditionally, CRP has been used clinically for the diagnosis and monitoring of autoimmune and infectous disorders. Recent studies have shown that chronic inflammation is an important component in the development and progression of atherosclerosis. As a result, increased serum CRP concentration are positively associated with the risk of future coronary events.
