    Principle:
    • This employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for human MIA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any human MIA present is bound by the immobilized antibody. After washing away any unbound substances, an enzyme-linked polyclonal antibody specific for human MIA is added to the wells. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of human MIA bound in the initial step. The color development is stopped and the intensity of the color is measured.

    Hintergrund

    MIA, also known as cartilage-derived retinoic acid-sensitive protein (CD-RAP), is a secreted protein expressed in cartilaginous tissue during development and in the adult. MIA was identified based on the fact that it is highly expressed in malignant melanomas, but not in normal melanocytes. MIA has been shown to interact with fibronectin, and promotes invasion and metastasis of melanoma cells, perhaps by blocking integrin-fibronectin interactions.
