One paralog of mammalian wisp2 was found in zebrafish.
CCN3 (Nov (zeige NOV Proteine)) and CCN5 (WISP2) are novel substrates of MMP14 (zeige MMP14 Proteine).
Results show that WISP2 and beta-catenin (zeige CTNNB1 Proteine) were more highly expressed in gastric cancer tissues and seem to correlate with early stage or without metastasis.
Studies indicate that the CYR61 (zeige CYR61 Proteine) CTGF (zeige CTGF Proteine) NOV (zeige NOV Proteine) matricellular proteins (CCN family of proteins) comprises the members CCN1 (zeige CCNA2 Proteine), CCN2 (zeige CTGF Proteine), CCN3 (zeige NOV Proteine), CCN4 (zeige WISP1 Proteine), CCN5 and CCN6 (zeige WISP3 Proteine) and have been identified in various types of cancer.
Report opposing effects of CCN2 (zeige CTGF Proteine) and CCN5 on fibroblast proliferation and transdifferentiation induced by TGF-beta (zeige TGFB1 Proteine).
Serum WISP2 correlated directly with fatty acid binding protein 4. Serum SFRP5 (zeige SFRP5 Proteine) did not differ between obese (n=32) vs. nonobese (n=25) PCOS women, but reference women had lower SFRP5 (zeige SFRP5 Proteine) (p<5x10(-6) as compared to both PCOS groups).
Activation of CCN5 may have the therapeutic potential to kill triple-negative breast cancer.
WISP2 has a role in regulating tumor cell susceptibility through EMT (zeige ITK Proteine) by inducing the TGF-beta (zeige TGFB1 Proteine) signaling pathway, KLF-4 (zeige KLF4 Proteine) expression and miR-7 (zeige LILRB1 Proteine) inhibition.
overexpression of FGFBP1 (zeige FGFBP1 Proteine) or loss of WISP-2 expression is closely related to the metastasis, invasion and poor prognosis of gallbladder cancer.
Loss of WISP2 in estrogen-dependent MCF7 human breast cancer cells promotes a stem-like cell phenotype.
We demonstrate that the overexpression of CCN5 in lung fibroblasts suppresses the upregulation in the expression of alpha-SMA (zeige SMN1 Proteine) and collagen induced by CCN2 (zeige CTGF Proteine).
Insulin (zeige INS Proteine)-like growth factor (IGF)-1 (zeige IGF1 Proteine) elicited clear stimulation of WNT1 inducible signaling pathway protein 2 (CCN5/WISP2) mRNA and protein levels in cultured islets.
Data suggest that CCN5, minimally expressed in unstimulated islet beta-cells, is strongly and directly induced by Igf1 (insulin-like growth factor I (zeige IGF1 Proteine)); overexpression of CCN5 stimulates cell proliferation and inhibits streptozotocin-induced apoptosis.
These results suggested that secreted Wisp2/CCN5 induces Akt (zeige AKT1 Proteine) and ERK (zeige EPHB2 Proteine) phosphorylation via integrins, and consequently facilitates neurite formation and conferred resistance to apoptosis.
CCN2 (zeige CTGF Proteine) is pro-hypertrophic and -fibrotic, whereas CCN5 is anti-hypertrophic and -fibrotic.
Although WISP-2 is a downstream protein of Wnt (zeige WNT2 Proteine) signaling, the role of WISP-2 on adipocyte differentiation may be marginal, at least in this in vitro culture model.
This gene encodes a member of the WNT1 inducible signaling pathway (WISP) protein subfamily, which belongs to the connective tissue growth factor (CTGF) family. WNT1 is a member of a family of cysteine-rich, glycosylated signaling proteins that mediate diverse developmental processes. The CTGF family members are characterized by four conserved cysteine-rich domains: insulin-like growth factor-binding domain, von Willebrand factor type C module, thrombospondin domain and C-terminal cystine knot-like (CT) domain. The encoded protein lacks the CT domain which is implicated in dimerization and heparin binding. It is 72% identical to the mouse protein at the amino acid level. This gene may be downstream in the WNT1 signaling pathway that is relevant to malignant transformation. Its expression in colon tumors is reduced while the other two WISP members are overexpressed in colon tumors. It is expressed at high levels in bone tissue, and may play an important role in modulating bone turnover.
