    Application Note: Anti-Streptavidin Peroxidase antibody has been tested by ELISA and western blot. This product has been assayed against 1.0 μg of Streptavidin in a standard capture ELISA using ABTS (2,2'-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) for 30 minutes at room temperature. Optimal concentrations in immunoassays should be determined by researcher. ELISA Dilution: 1:2,000-1:10,000

    Store vial at -20° C or below prior to opening. This vial contains a relatively low volume of reagent (25 μL). To minimize loss of volume dilute 1:10 by adding 225 μL of the buffer stated above directly to the vial. Recap, mix thoroughly and briefly centrifuge to collect the volume at the bottom of the vial. Use this intermediate dilution when calculating final dilutions as recommended below. Store the vial at -20°C or below after dilution. Avoid cycles of freezing and thawing.

    Hintergrund

    Background: Anti-streptavidin antibody detects Streptavidin, a 60 kDa protein purified from the bacterium Streptomyces avidinii. Streptavidin is a homo-tetrameric protein having a very high affinity for biotin (vitamin B7). Streptavidin bound biotin has a dissociation constant (Kd) on the order of about 10^-14 Mol/L. Streptavidin and biotin are used extensively in molecular biology and bionanotechnology due to the resistance of streptavidin-biotin complex to organic solvents, denaturants (e.g. guanidinium chloride), detergents (e.g. SDS, Triton), proteolytic enzymes, and extremes of temperature and pH .
