    Applikation
Immunodiffusion (ID), Immunoelectrophoresis (IEP), Nephelometry (Neph), Radial Immunodiffusion (RID), ELISA
    Spezifität
The antiserum does not cross-react with any other component of mouse plasma. Inter-species cross-reactivity is a normal feature of antibodies to plasma proteins since they frequently share antigenic determinants. of this antiserum has not been tested in detail.
    Produktmerkmale
Precipitating polyclonal goat antiserum to mouse major urinary proteins
    Aufreinigung
Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies cross-reacting with other with other proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.
    Immunogen
The MUP is a sex related complex with a pre-albumin mobility in gel electrophoresis. Concentrations in male mice are between 50 to 150 µ g/ml, in female mice 10 to 20 µ g/ml. The complex is also detectable in serum, however concentrations are about 200 to 400 times lower. Variations in the composition of the complex are genetically determined. Purified mouse major urinary proteins are isolated from pooled normal urine. Freund’s complete adjuvant is used in the first step of the immunization procedure.
    Isotyp
    IgG
  • Applikationshinweise
In precipitating techniques as immunoelectrophoresis and single or double radial immunodiffusion (Mancini, Ouchterlony) and electroimmunodiffusion (Laurell) to identify or measure the concentration of MUP. Proteins isolated from urine, such as Bence Jones proteins, may contain MUP as an impurity. This antiserum can be used to identify the impurity and also to remove it by immunoaffinity adsorption. It has not been tested for use in nephelometry, ELISA or immunohistology, but this does not exclude such use if proper controls are included.
    Beschränkungen
    Nur für Forschungszwecke einsetzbar
  • Format
    Lyophilized
    Konzentration
Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal goat serum. No foreign proteins added. Antibody titre: Precipitin titre 1:32 when tested against pooled normal mouse urine in agar-block immunodiffusion titrati
    Buffer
Delipidated, heat inactivated, lyophilized, stable whole antiserum
    Konservierungsmittel
    Without preservative
    Lagerung
    4 °C/-20 °C
    Informationen zur Lagerung
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C, prolonged storage at or below -20°C. Reconstitute the lyophilized antiserum by add ing 1 ml sterile distilled water. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at +4°C, not ref rozen, and preferably used the same day.
  • Target
    Major Urinary Proteins
    Hintergrund
The reactivity of the antiserum is restricted to mouse MUP. In immunoelectrophoresis and radial immunodiffusion, using various antiserum concentrations against normal mouse plasma characteristic precipitin lines are obtained which show a reaction of identity with the precipitin lines obtained with purified immunogen. No reaction is obtained with any other plasma protein components or serum
