The Yes1-selective antibodies are affinity purified on an immobilized antigen based affinity matrix, the isolated antibodies were then stabilized in antibody stabilization buffer for long-term storage. The Yes1-selective antibodies are fully characterized for applications in western blotting and ELISA at the recommended dilutions. The Supplier provides Yes1 Western blot positive control samples in SDS-PAGE sample buffer.

Protein tyrosine kinases are large multigene families which are important mediators of the signaling cascade, responsible for intracellular signal transduction. Tyrosine kinases are a diverse group of cell surface transmembrane proteins that act as receptors for cytokines, growth factors, hormones and other signaling molecules. Receptor tyrosine kinases are expressed in many cell types and play important roles in a wide variety of cellular processes, including growth, differentiation, angiogenesis, metabolism and apoptosis in response to external and internal stimuli. Over-expression and/or structural alteration of tyrosine kinase family members are associated to human cancers. Tumor cells use RTK transduction pathways to achieve tumor growth, angiogenesis and metastasis making it a target in approaches of cancer therapy. Yes1 kinase, a non-receptor protein kinase, is one of the 9 members of the Src family kinase (SFK). Yes 1 kinase is the cellular homolog of the Yamaguchi sarcoma virus oncogene that has tyrosine kinase activity and is ubiquitously expressed in many tissues and cells. It contains several conserved functional domains such as an N-terminal myristoylation sequence for membrane targeting, SH2 and SH3 domains, a kinase domain, and a C-terminal non-catalytic domain. It lies in close proximity to thymidylate synthase gene on chromosome 18 and 22, and its activation plays a significant role in the malignant transformation of hepatocytes. The Src kinase Yes is activated in pancreatic acinar cells by gastrointestinal hormones/neurotransmitters, but not pancreatic growth factors, which stimulate its association with many other signaling molecules. Thus, Yes1 is a useful marker to detect early-stage hepatocellular carcinoma, and plays a key role in the tumorigenesis and metastasis of gastric cancer. Yes1 induction results in increased cancer cell motility thus promoting cancer spread and metastasis rather than tumor growth. Yes 1 is involved in protein sorting or vesicular trafficking due to its being the sole SFK present in lipid rafts enriched in caveolin in MDCK cells. YES1 downregulation has a significant effect on the survival of basal-like breast cancer cell lines. Thus, Yes1 kinase is a therapeutic target in a number of cancers including melanomas, breast cancers, and rhabdomyosarcomas.The gene for YES1 is present on chromosome 18p11.31-p11.21.
