Hintergrund: Ets-Related Gene (ERG) product is a ~58 kDa nuclear protein which functions as a DNA-binding transcriptional regulator and belongs to the erythroblast transformation-specific (ETS) family of transcription factors. It is a nuclear and cytoplasmic transcriptional regulator that undergoes alternative splicing to produce multiple isoforms, designated Erg-1, Erg-2 and Erg-3. ERG plays an important role in cell development, differentiation,proliferation, apoptosis and tissue remodeling.This family of transcription factors contains approximately 30 members that share a highly conserved DNA-binding domain (ETS domain) and differs from each other in other domains (such as absence or presence of the Pointed/SAM domain). ERG is linked to normal processes such as mesoderm formation and is found to form functional complexes with Jun/Fos, with the resulting ternary complexes regulating expression of proteins such as metalloprotease-1 (MMP-1) and MMP-3. ERG protein promotes the transcriptional activation of oncogenes and facilitates cancer progression. ERG is associated with prostatic intraepithelial neoplasia (PIN) and benign and malignant vascular endothelial cells in various different vascular tumors. ERG oncoprotein expression is a highly specific marker for prostate cancer as it is present in 50-70 % of all prostate carcinomas. Defects in ERG are a cause of Ewing sarcoma which is a highly malignant, metastatic, primitive small round cell tumor of bone and soft tissue that affects children and adolescents. Obesity is linked with poorer prostate cancer prognosis primarily in men with tumors harboring the gene fusion TMPRSS2: ERG. The human ERG gene maps to chromosome 21 and encodes a 486 amino acid protein.
