Chain of chicken CD8 (Mr 34 kDa) In the chicken, the CD8 molecule is present in two forms: (i) a homodimer of two chains, and (ii) a heterodimer of an chain and a chain.1,2 Chicken CD8 is expressed on approximately 80% of thymocytes, 45% of blood mononuclear cells and 50% of spleen cells, but <1% of cells in the bursa and bone marrow.1 While the vast majority of CD8+ cells in the thymus, spleen, and blood of adult chickens express both CD8- and CD8-chains, a relatively large proportion of the CD8+ TCR- cells in the spleens of embryos and young chicks express only the -chain of CD8. Among intestinal epithelial lymphocytes, the major CD8+ T cell populations present in mice are conserved, but there is a population of TCR- CD8 cells in the chicken that is not found in rodents.3 MAb 3-298 recognizes the CD8 chain of both the chicken and turkey.

Working Dilutions: Flow Cytometry: Fluorescein (1 ug/10^6 cells), Biotin (1 ug/10^6 cells), R-phycoerythrin (0.2 ug/10^6 cells), Cyanine 5 (1 ug/10^6 cells). Since applications vary, you should determine the optimum working dilution of the product that is appropriate for your specific need.

The purified (UNLB) antibody is supplied as 0.5 mg of purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.0. No preservatives or amine-containing buffer salts added. Store at 2-8C. The fluorescein (FITC) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8C. The biotin (BIOT) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8C. The R-phycoerythrin (R-PE) conjugate is supplied as 0.1 mg in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8C. Do not freeze! The Cyanine 5 (CYTM5) conjugate is supplied as 0.1 mg in 1.0 mL of PBS/NaN3. Store at 2-8C. Protect conjugated forms from light. Each reagent is stable for the period shown on the bottle label if stored as directed. WARNING Reagents contain sodium azide. Sodium azide is very toxic if ingested or inhaled. Avoid contact with skin, eyes, or clothing. Wear eye or face protection when handling. If skin or eye contact occurs, wash with copious amounts of water. If ingested or inhaled, contact a physician immediately. Sodium azide yields toxic hydrazoic acid under acidic conditions. Dilute azide-containing compounds in running water before discarding to avoid accumulation of potentially explosive deposits in lead or copper plumbing.
