Beschreibung C1q is a sub-component of the first complement component C1, which is formed by theinteraction of C1q with the tetramer C1s-C1r-C1r-C1s. C1q is a member of a family of soluble collagen-like proteins and is composed of threebasic subunits termed A, B and C (27,550, 25,200, 23,800 Da respectively) which asociatein the proportion 6 A-B subunits (52, 750 Da) and 3 C-C subunits (47,600 Da), to formwhole C1q (459,300 Da). C1q binds to IgG and IgM immune complexes, this activates C1rand C1s, which are also bound to C1q and so initiates the classical pathway of complementactivation.
Spezifität Detection of C1q in serum, in serum-C1 and within immune complexes, inhibits hemolyticactivity of C1 by binding to C1q in the liquid phase (antibody concentration necessary toinhibit 50% of hemolytic activity in the fluid phase is 0. 85 µg/ml). With C1 bound toimmune complexes a dose dependent increase of C1 activity was observed at antibodyconcentrations of 1. 5 µg/ml (cf. ref. 1). Useful for detection of C1q-bearing immune complexes in patients with rheumaticdisorders. Polypetide Reacting: C-chain of complement C1q (25 kD in SDS-PAGE). Species: Human. Others not tested.

Anwendungen

Applikationshinweise ELISA. Inhibition experiments (e. g. hemolytic activity). Immunoblotting (Western): 1/100, 1h at RT (ECL method). Other applications not tested. Optimal dilutions are dependent on conditions and should be determined by the user.
Buffer Final Solution contains PBS buffer, pH 7.4 with 0.09% Sodium Azide aspreservative and 0.5% BSA as stabilizer.Reconstitution: Restore in 1 ml distilled water.
Lagerung Store the antibody at 2-8°C. Shelf life: one year from despatch.
