Immunogen: This affinity purified antibody was prepared fromwhole rabbit serum produced by repeated immunizations witha synthetic peptide corresponding to aa 115-129 (of 273) ofyeast DNA Replication Factor A (RFA2). The RFA complexconsists of a heterotrimer of RFA1, RFA2 and RFA3, 69, 36and 13 kda chains respectively. Yeast RFA2 isphosphorylated at the G 1 /S transition and is dephosphorylated at mitosis. Spezifität: This affinity purified antibody isdirected against yeast RFA2. The product was affinity purifiedfrom monospecific antiserum by immunoaffinity purification.Antiserum was first purified against the phosphorylated form ofthe immunizing peptide. The resultant affinity purified antibodywas then cross-adsorbed against the non-phosphorylated formof the immunizing peptide. This phospho specific polyclonalantibody is specific for phosphorylated pS122 of yeast RFA2.Reactivity with non-phosphorylated yeast RFA2 is minimal byELISA.

Anwendungen

Applikationshinweise: Recommended Dilutions: This product was assayed by western blotting against RFA2 containing cell lysates. A 1:500 to1:1000 dilution is recommended for this assay. This product was also assayed by ELISA against 0.1 ug of the phosphorylatedform of the immunizing peptide. A 1:8000 to 1:20000 dilution of the antibody is recommended for this assay. Equivalentsignal is seen against the non-phosphorylated form of the peptide when the antibody is diluted to 1:300. Note: This affinity-purified antibody was tested for use in western blotting against RFA2 containing cell lysatesand by ELISA against the immunizing peptide. Reactivity in other immunoassays is unknown.
