ADAM10, as known as disintegrin and metalloproteinase domain-containing protein 10 isoform 1, is a member of the ADAM family that contains a disintegrin and metallogrotease-like domain. This protein is a sheddase, and has a broad specificity for peptide hydrolysis reactions. Also, it cleaves ephrin, within the ephrin/eph complex, formed between two cell surfaces. When ephrin is freed from the opposing cell, the entire ephrin/eph complex is endocytosed. In neurons, ADAM10 is the most important enzyme with alpha-secretase activity for proteolytic processing of the amyloid precursor protein. Recombinant human ADAM10, fused to His-tag at C-terminus, was expressed in insect cell and purified by using conventional chromatography techniques.
