Beschreibung KLRG1 (Mr 30-38 kDa) The monoclonal antibody 2F1 reacts with mouse killer cell lectin-like receptor G1 (KLRG1, formerly known as mouse MAFA or 2F1-Ag), a homodimeric member of the lectin-like type 2 transmembrane receptor family that contain characteristic immunoreceptor tyrosine-based inhibitory motifs (ITIMs) in their cytoplasmic domains.1, 2 These ITIMs interact with the SH2 domains of protein phosphatases such as SHP-1.3, 4 The 2F1 antibody stains 30-60% of NK1.1+CD3- splenocytes, and a small fraction of T cells in all mouse strains tested (C57BL/6, BALB/c, 129/J, C3H.SW, AKR/J, SJL).1, 5 Cell surface expression of KLRG1 is up-regulated by expression of MHC class I molecules. The effect of MHC class I expression is indirect, and can be mediated by interactions with class I-specific Ly49 inhibitory receptors.

Anwendungen

Applikationshinweise Working Dilutions: Flow Cytometry: Purified (1 ug/10^6 cells), Fluorescein (1 ug/10^6 cells), Biotin (1 ug/10^6 cells), R-phycoerythrin (0.2 ug/10^6 cells). Since applications vary, investigators are advised to determine the optimum working dilution of the product that is appropriate for a specific application.
Lagerung The purified (UNLB) antibody is supplied as 0.5 mg of purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.0. No preservatives or amine-containing buffer salts added. Store at 2-8C. The fluorescein (FITC) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8C. The biotin (BIOT) conjugate is supplied as 0.5 mg in 1.0 mL of PBS/NaN3. Store at 2-8C. The R-phycoerythrin (R-PE) conjugate is supplied as 0.1 mg in 1.0 mL or 0.2 mg in 2.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8C. Do not freeze! The low endotoxin, azide-free (LE/AF) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mL of PBS. Aliquot and store at or below -20C. Protect conjugated forms from light. Aliquot and freeze the low endotoxin, azide-free product at -20C immediately upon receipt. Each reagent is stable for the period shown on the bottle label if stored as directed. WARNING Reagents contain sodium azide. Sodium azide is very toxic if ingested or inhaled. Avoid contact with skin, eyes, or clothing. Wear eye or face protection when handling. If skin or eye contact occurs, wash with copious amounts of water. If ingested or inhaled, contact a physician immediately. Sodium azide yields toxic hydrazoic acid under acidic conditions. Dilute azide-containing compounds in running water before discarding to avoid accumulation of potentially explosive deposits in lead or copper plumbing.
