Immunogen: This protein A purified monoclonal antibody was produced by repeated immunizations with purifiedhuman cytoskeletal preparations from A431 cells. Hybridoma: Produced by the fusion between BALB/c mouse splenocytes and mouse myeloma SP2/0 cells afterimmunization with cytoskeletal preparations from human A431 cells using conventional hybridoma technology.

Beschreibung: Cytokeratins(CK) are intermediate filaments of epithelialcells, both in keratinizing tissue (i.e. skin)and non-keratinizing cells (i.e. mesothelial cells). Although not a traditional marker for endothelial cells, cytokeratins have also been found in some microvascularendothelial cells. At least 20 different cytokeratins (CK) in the molecular range of 40-70 kDa and isoelectric points of 5-8.5 can be identified using two dimensional gel electrophoresis. Biochemically, most members of the CK family fall intoone of two classes, type I (acidic polypeptides) or type II (basic polypeptides). At least one member of both the acidicand basic families are expressed in epithelial cells. Monoclonal antibodies to cytokeratin proteins can be usefulmarkers for tumor identification and classification. Anti-Keratin (pan) reacts with a variety of keratins (4, 5,6,8,10,13 and18) in a variety of normal and neoplastic epithelial. This antibody is also useful to stain cultured epithelial cells. Thisantibody reacts with simple epithelium and both basal and superbasal layers of squamous epithelium. Anti-keratin (pan)is useful to differentiate epithelial tumors from non-epithelial tumors. Keratins have a cytoplasmic localization.

Spezifität: This protein A purified mouse monoclonal antibody reacts specifically with keratins from humantissues and derived cell lines. This antibody reacts with keratin (56 kDa), keratin 17 (46 kDa), keratin 18 (45 kDa) andkeratin 19 (40 kDa) derived from humans. Cross reactivity with keratins from other sources has not been determined.No reaction is expected against other filament proteins including vimentin, desmin and neurofilament protein.

Lagerung: Store vial at -20° C prior to opening. Aliquot contents and freeze at -20° C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use. Expiration date is one (1) year from date of opening.

Western blot using antibodies-online Immunochemical's Mouse Mab-anti-Keratin antibody. This antibody recognizes a single 56 kDa band corresponding to human keratin as confirmed by the position of molecular weight markers (not shown). Approximatley 100 ng of keratin from human epidermis (Sigma p/n K0253) was applied under reducing conditions to a pre-cast 4-20% iGel from Gradipore Inc.
