Western Blotting (WB), Immunoprecipitation (IP), ELISA
    Spezifität
The antibody labels a strong band of CD4 at 55 kDa in ABIN1741637 samples & in other cancer cell lines.
The antibody also labels a band of 28 kDa, the identity ofthis band is not fully understood but may represent a proteolytic degradation product of CD4 in SKNMC cells.
    Kreuzreaktivität
    Maus, Ratte (Rattus), Affe
    Kreuzreaktivität (Details)
Does not cross react to any other cellular protein.
    Produktmerkmale
CD4 selective antibodies were generated against a peptide taken from the human protein. The CD4 selective antibodies are affinity purified on an immobilized antigen based affinity matrix, the isolated antibodies were then stabilized in antibody stabilization buffer for long-term storage. The CD4-selective antibodies are fully characterized for applications in western blotting and ELISA at the recommended dilutions. The supplier provides CD4 Western blot positive control samples in ready-to-use SDS-PAGE sample buffer. Limited quantities of antigenic blocking peptide for CD4 antibody is also available, please inquire before ordering.

    Hintergrund
Human T-cell surface glycoprotein CD4, also known as T-cell surface antigen T4/Leu-3, is a single chain trans membranous glycoprotein (55 kDa) which belongs to the immunoglobulin superfamily. CD4 (Cluster of Differentiation 4) consists of a cytoplasmic tail, four immunoglobulin-like external domains (D1-D4), and a transmembrane domain. CD4 contains three Ig-like C2-type (immunoglobulin-like) domains and one Ig-like V-type (immunoglobulin-like) domain. CD4 is present on a subset of T lymphocytes (helper/inducer T cells) and at a lower level on monocytes, regulatory T cells, tissue macrophages, granulocytes, Langerhans cells and dendritic cells. It is also expressed in specific regions of the brain. It is an important component of the immune system and is the cellular receptor for HIV-1. This protein initiates the early phase of T-cell activation, and is an important mediator of indirect neuronal damage in infectious and immune-mediated diseases of the central nervous system. CD4 plays a role in the regulation of T-B lymphocyte adhesion in the absence of antigen recognition, and in thymic differentiation. Subpopulations of suppressor or cytotoxic T cells are identified among CD4-positive T cells. The intracellular domain of the antigen is associated with p56 lck protein tyrosine kinase. CD4 is a co-receptor that assists the T cell receptor (TCR) to activate its T cell following an interaction with an antigen presenting cell. Using its portion that resides inside the T cell, CD4 amplifies the signal generated by the TCR. The antigen interacts directly with major histocompatibility complex (MHC) class II molecules on the surface of the antigen presenting cell via its extracellular domain. It is also a major receptor for the human immunodeficiency virus (HIV), binding directly to the envelope glycoprotein gp120 on HIV. HIV infection leads to a progressive reduction of the number of T cells possessing CD4 receptors thus making the CD4 count an important factor to decide when to begin treatment for HIV-infected patients. The human CD4-specific gene maps to chromosome 12pter-p12.
