Structural basis of the interaction between TOP3B and the TDRD3 auxiliary factor has been reported.
Results identified TOP3B gene deletion as candidate gene in the etiology of neurodevelopmental disorders including juvenile myoclonic epilepsy.
Top3beta mutants lacking either its RNA-binding domain or catalytic residue fail to promote synapse formation, suggesting that Top3beta requires both its mRNA-binding and catalytic activity to facilitate neurodevelopment.
Top3b proteins from several animals associate with polyribosomes, which are units of mRNA translation, whereas the Top3 homologs from E. coli and yeast lack the association. The Top3b-polyribosome association requires TDRD3, which directly interacts with Top3beta and is present in animals but not bacteria or yeast.
TOP3B promotes transcription, protects against DNA damage, and reduces the frequency of chromosomal translocations.
Topoisomerase IIIbeta expression can be a useful marker in assessing the prognosis of patients with breast cancer and is an independent predictor of survival.
the positive regulatory region including the 5'GGAACC3' core element is essential for efficient expression of the DNA topoisomerase III beta gene
Two isoforms of human DNA topoisomerase IIIbeta were expressed in HeLa cells. Isoform 1 was localized to the nucleus and associated with chromosomes during metaphase and anaphase. Isoform 2 was found in the cytoplasm.
This study indicated a role for TOP3beta in mRNA metabolism and suggest that it is involved in neurodevelopmental disorders.
DNA topoisomerase III beta has a role in infertility and aneuploidy
Chromosomal abnormalities in top3beta(-/-) mice might lead to a persistent increase in apoptotic cells, which might in turn lead to the progression of autoimmunity
suggest an unanticipated role for Top3beta beyond DNA repair in the activation of cellular responses to DNA damage
This gene encodes a DNA topoisomerase, an enzyme that controls and alters the topologic states of DNA during transcription. This enzyme catalyzes the transient breaking and rejoining of a single strand of DNA which allows the strands to pass through one another, thus relaxing the supercoils and altering the topology of DNA. The enzyme interacts with DNA helicase SGS1 and plays a role in DNA recombination, cellular aging and maintenance of genome stability. Alternative splicing of the C-terminus of this gene results in three transcript variants which have distinct tissue specificity\; however, not all variants have been fully described.
