Hintergrund Ionotropic Glutamate Receptor 2 (GluR2) is a predominant excitatory neurotransmitter in mammalian brain located at human chromosome 4q32-33 (1). It belongs to a family of alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionate (AMPA) receptors (2). GluR2 plays a crucial role in controlling the calcium permeability of AMPA receptors and therefore, its presence or absence greatly influences AMPA receptor properties and synaptic functions. GluR2 mRNA ordinarily undergoes post-transcriptional editing which is essential for brain function. AMPA receptors containing edited GluR2 have low conductance, low calcium permeability and confer a linear current-voltage (I-V) relationship. AMPA receptors lacking an edited GluR2 subunit have high calcium permeability and also exhibit inward rectifying I-V relationship (3). Defective GluR2 RNA editing might also play a role in the development of amyotrophic lateral sclerosis (ALS) (4). GluR2 is among the most common and abundant receptor in striatum and projection neurons. In rats, all striatal projection neurons express GluR2 (5). It is highly concentrated on the postsynaptic neuronal membranes of the superficial dorsal horn and its removal might contribute to spinal central sensitization in persistent inflammatory pain (6). It is expressed in cortex, hippocampus and cerebellum.
