Hintergrund Caspase-3 has been extensively studied and implicated to play an important role in apoptosis. Bewertung The antibody works for IHC on frozen sections. Among the dilutions tested, 1:20 gave the best results. Both 1:10 and 1:50 dilutions gave a weaker signal respect to 1:20. The signal for Casp3 was detected both in cerebral cortex and cerebellum.

Protokoll
  • Mouse cerebellum at post natal day (P) 5 was perfusion fixed in paraformaldehyde 4% in 0.1M phosphate buffered saline (PBS) pH7.4 and post-fixed in the same fixative for 4 additional hours.
  • Following several washes in PBS, cerebellum was cryoprotected in 30% sucrose, frozen in liquid nitrogen and cut in 30μm thick sections.
  • Sections were rinsed in PBS for about 30min.
  • Sections were blocked in 0.01M PBS containing 0.1% Triton and 5% Normal Goat Serum (NGS) for 1h at RT.
  • Incubation with primary antibody ABIN222873 diluted 1:10/1:20/1:50 in the blocking solution ON at 4°C. The negative control slice was incubate with blocking solution only ON at 4°C.
  • Three 5min washed in 0.01M PBS at RT.
  • Incubation with the anti-rabbit secondary antibody (Invitrogen, AF488, Lot 1073084) diluted 1:800 in 0.1M PBS-0.1% Triton for 1h at RT.
  • Three 5min washed in 0.01M PBS at RT.
  • Specimens were mounted in Fluoroshield mounting medium (Sigma, F6182, Lot MKBX3783V).
  • Images were taken on a Leica SP5 upright confocal microscope (Magnification 40x, zoom 1.47).
Anmerkungen In order to exclude any nonspecific signal due to secondary antibody, for each experimental procedure, one control slice (negative control) was processed omitting the primary antibody: in this case the overnight incubation at 4°C degree was performed in diluent solution only.
