Immunogen This whole rabbit antiserum was prepared by repeated immunizations with a GST fusion protein. The epitope maps near the carboxy-terminus of human Erk-2. The epitope is identical to the corresponding sequence in mouse and differs from the rat sequence by a single, conservative amino acid substitution.

Beschreibung Cell proliferation is regulated in several contexts, for example during development, tissue differentiation, wound healing and immune responses. In mammalian cells, proliferative signals lead to the activation of a protein kinase cascade, resulting in the phosphorylation of two closely related Mitogen-Activated Protein Kinases (MAPKs) ERK1 and ERK2. When activated, ERKs form dimers that translocate to the nucleus where they phosphorylate several classes of transcription factors which are involved in the up-regulation of immediate early genes. As such, Erk1 and Erk2 represent a paradigm for a growing family of proline-directed protein kinases that mediate entry, progression and exit from the cell cycle in diverse eukaryotic cells. These enzymes function within highly conserved cascade of sequentially activating protein kinases that transduce signals from diverse extracellular stimuli. In vertebrates the mitogen-induced sequential activation of the kinases Raf1->Mek1->Erk2->Rsk occurs via the G-protein Ras.
Spezifität This antiserum is directed against Erk-2 and is useful in determining its presence in immunoblotting experiments. This antibody does not cross react with Erk-1. This antibody reacts with Erk-2 in human and mouse. Reactivity to rat tissues is anticipated due to sequence similarities between the peptide antigen and the corresponding sequence found in rat.

Anwendungen

Applikationshinweise Note: This antiserum recognizes Erk-2 (extracellular signal-regulated kinase 2) from human and mouse sources. Erk-2 is a ubiquitous protein kinase target for Ras and Raf. The following cell lines have been assayed by immunoblot and were found to be positive for Erk-2 using this reagent: U937, HeLa, NIH-3T3, RAW 264.7, LNCaP and HEK whole cell lysates. Reactivity with Erk-2 from other sources is unknown but likely.

Lagerung Store vial at -20° C prior to opening. Aliquot contents and freeze at -20° C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4° C as an undiluted liquid. Dilute only prior to immediate use. Expiration date is one (1) year from date of opening.
