Hintergrund Muscular hypertrophy is generally associated with extended period of passive muscle stretch which is associated with increased mRNA and protein synthesis. There are several hypertrophic agonist have been identified as probable candidates for such muscle anabolic effects. However, little is known as to how the mechanical exercise is converted in to intracellular signals regulating gene expression resulting in an increase in cell size and is regarded as a beneficial adaptive response to increased exercise/workload, but in some instances hypertrophy can be detrimental as seen in long-term cardiac hypertrophy. On the other hand, muscle wasting, muscle atrophy, is a feature of both inherited and acquired muscle disease and also as normal aging processes. Certain genes were identified by substractive mRNA hybridization prepared from passively stretched and control murine skeletal muscle. One of the mRNA that undergoes increased expression in response to passive stretch codes for one RNA that is significantly increased in passively stretched muscle. Encoded within the mRNA is an open reading frame of 311 amino acids (38 kDa) containing a highly conserved type 1 peroxisomal targeting signal and a serine lipase active center. The sequence shows identity to a family of serine hydrolases and thus is named serine hydrolase-like (Serhl) or karaken-like. There are 2 SERHL variants identified, SERHL1 and SERHL2, both are ubiquitously present, in higher amounts in kidney and skeletal muscle. The predicted three-dimensional structure displays a core alpha/beta-hydrolase fold and catalytic triad characteristic of several hydrolytic enzymes. SERHL is located on human chromosome 22q13 (2) encoded by 5 exons and most likely have splice variants. This protein is a cytoplasic protein that is concentrated in perinuclear vesicvles and based on sequence similarity may be associated with peoxisomes. SERHL1 and SERHL2 are believed to play an important role in skeletal muscle development and peroxisome function in response to mechanical stimuli. Both SERHL1 and SERHL2 are ubiquitously present, high expression is noted in kidney, skeletal muscle and large intestine.
