Beschreibung The BCL10 gene encodes a 233 amino acid protein with an N-terminal caspase recruitment domain (CARD), found in a number of apoptotic regulatory molecules. Initial in vitro transient overexpression experiments suggest that BCL10 induces apoptosis and activates nuclear factor- κΒ (NF-κΒ) (1, 2). Translocations involving the BCL10 gene are rare in MALT lymphomas, accounting for fewer than 5% of all cases, and so far only in MALT lymphomas arising in the stomach and the lungs. The t(1,14) translocation juxtaposes the BCL10 gene next to the immunoglobulin heavy chain gene, and leads to overexpression and aberrant nuclear localization of the BCL10 protein. Virtually all cases of MALT lymphoma carrying the t(11,18) translocation also show aberrant nuclear expression of BCL10, albeit at much lower levels. Mutations of the BCL10 gene have been reported in a small proportion of B-cell lymphomas including MALT lymphomas. The mutations often lead to expression of a truncated BCL10 protein, however, retaining the CARD domain which is sufficient and necessary for NF- κΒ activation (2).
Spezifität In Western blotting of homogenized frozen tissue from tonsil, MALT-lymphoma and HEK 293 cells, the antibody predominantly labels a band of 32 kDa and a weaker band of 37 kDa (1). Western blotting of homogenized HEK 293 cells transfected with different BCL10 products indicates that the antibody recognizes an epitope between amino acids 122-168 (1).

Anwendungen

Applikationshinweise Paraffin sections: The antibody can be used for labelling paraffin-embedded tissue sections fixed in formalin. Pre-treatment of tissues with heat-induced epitope retrieval is required. Optimal results are obtained with DakoCytomation Target Retrieval Solution, High pH, code No. S 3308, or DakoCytomation Target Retrieval Solution, pH 9, code No. S 2368. Less optimal results are obtained with DakoCytomation Target Retrieval Solution, code No. S 1700, or 10 mmol/L citrate buffer, pH 6.0. Pre-treatment of tissues with DakoCytomation Proteinase K, code No. S 3020, was found inefficient. The tissue sections should not dry out during the treatment or during the following immunocytochemical staining procedure. Dilution: Monoclonal Mouse Anti-Human BCL10 Protein, code No. M 7260, may be used at a dilution range of 1:25-1:50 when applied on formalin-fixed, paraffin-embedded sections of human tonsil or MALT lymphomas with t(1,14) translocation and using 20 minutes heat-induced epitope retrieval in DakoCytomation Target Retrieval Solution, pH 9, code No. S 2368, and 30 minutes incubation at room temperature with the primary antibody. Optimal conditions may vary depending on specimen and preparation method, and should be determined by each individual laboratory. The recommended negative control is DakoCytomation Mouse IgG1, code No. X 0931, diluted to the same mouse IgG concentration as the primary antibody. Unless the stability of the diluted antibody and negative control has been established in the actual staining procedure, it is recommended to dilute these reagents immediately before use, or dilute in DakoCytomation Antibody Diluent, code No. S 0809. Positive and negative controls should be run simultaneously with patient specimen. Visualization: DAKO LSAB/ HRP kit, code No. K 0679, and DAKO EnVision/HRP kits, code Nos. K 4004 and K 4006, are recommended. Follow the procedure enclosed with the selected visualization kit. Automation: The antibody is well-suited for immunocytochemical staining using automated platforms, such as the DakoCytomation Autostainer. Performance characteristics: Cells labelled by the antibody display staining confined to the cytoplasm, but in some cases the nucleus is labelled as well (1, 4). Normal tissues: The antibody labels normal spleen, thymus, lymph node, reactive tonsil and MALT of the appendix. BCL10 protein positive cells are both B and T cells with the majority of positive cells being T cells. Of non-lymphoid tissue the antibody labels luminal epithelial cells in breast. No labelling is observed in the following tissues: tongue, esophageal, duodenal, rectal, liver, gall bladder, pancreas, bronchus, lung, thyroid, adrenal gland, kidney, bladder, cervix, uterus, ovary and tube, placenta and cord, testis, heart, and skin (1). Cytoplasmic labelling is observed in tissues with high proliferation, e.g. germinal centre in tonsil, columnar epithelium in colon, kidney epithelium, and peripheral lymphocytes, and in addition, nuclear labelling in scattered cells especially in colon epithelium.
Buffer Monoclonal mouse antibody provided in liquid form as cell culture supernatant dialysed against 0.05 mol/L Tris/HCl, pH 7.2, and containing 15 mmol/L NaN 3 .

At product concentrations, though not classified as hazardous, sodium azide may react with lead and copper plumbing to form highly explosive build-ups of metal azides. Upon disposal, flush with large volumes of water to prevent metal azide build-up in plumbing. 3. As with any product derived from biological sources, proper handling procedures should be used.
