Beschreibung Syndecan-1 (1). CD138 est un protéoglycane transmembranaire, qui présente des sulfates d’héparan et parfois des sulfates de chondroitine. Il se compose d’une protéine centrale de 30,5 kDa et de cinq glycosaminoglycanes. CD138 appartient à la famille des syndecan. La masse moléculaire rapportée de CD138 va de 85 000 to 92 000, et dépend probablement du motif glycosaminoglycan de la molécule (1). Au sein du système hématopoïétique, CD138 se confine principalement aux stades de différenciations des lymphocytes-B. Il est généralement exprimé à forte densité cellulaire dans les cellules plasmatiques malignes (5,6), et dans les cellules lymphoplasmocytoïdes (6). Dans les tissus matures, CD138 est exprimé sur les épithélia simples et stratifiés, les fibroblastes, les kératinocytes stratifiés et cellules endothéliales (1). Spezifität Anti-CD138, MI15, was included in the Sixth International Workshop and Conference on Human Leucocyte Differentiation Antigens (Kobe 1996), and studies by a number of different laboratories confirmed its reactivity with CD138 (1). The epitope recognized by anti-CD138, MI15, is found within the ectodomain (extracellular part) of the CD138 core protein. Blocking experiments have revealed that the epitope recognized by MI15 seems to be partly overlapping the epitope recognized by another antibody to CD138, clone B-B4 (1, 5). Anti-CD138, MI15, reacts strongly with CD138 expressed by multiple myeloma-derived cell lines, such as U266 and XG-1 (9).

Anwendungen

Applikationshinweise Paraffin sections: The antibody can be used on formalin-fixed, paraffin-embedded tissue sections. Antigen retrieval by heating in DakoCytomation Target Retrieval Solution, code No. S 1700 or S 1699 is mandatory. Frozen sections and cell preparations: The antibody can be used for labelling acetone-fixed, frozen tissue sections, fixed cell smears, and fixed cytospins. The tissue sections should not dry out during the treatment or during the following immunocytochemical staining procedure. Dilution: Monoclonal Mouse Anti-Human CD138, code No. M 7228, may be used at a dilution range of 1:25-1:50 when applied on paraffin sections, acetone-fixed frozen tissue sections, fixed cell smears, and fixed cytospins, using 30 minutes incubation at room temperature with the primary antibody. Optimal conditions may vary depending on specimen and preparation method, and should be determined by each individual laboratory. The recommended negative control is DakoCytomation Mouse IgG1, code No. X 0931 diluted to the same mouse IgG concentration as the primary antibody. Unless the stability of the diluted antibody and negative control has been established in the actual staining procedure, it is recommended to dilute these reagents immediately before use, or dilute in DakoCytomation Antibody Diluent, code No. S 0809. Positive and negative controls should be run simultaneously with patient specimen. Visualization: DAKO LSAB/HRP kit, code No. K 0679, and DAKO EnVision/HRP kits, code Nos. K 4004 and K 4006, are recommended. For frozen sections and cell preparations, the DakoCytomation APAAP kit, code No. K 0670, is a good alternative if endogenous peroxidase staining is a concern. Follow the procedure enclosed with the selected visualization kit. Limitations Owing to the sensitivity of CD138 towards proteolytic cleavage, the processing of samples, such as freezing or storage before staining with the antibody, may play an important role in the outcome of the staining (1). Cells labelled by the antibody display staining confined to the cell surface membrane. In blood: No significant reaction is seen with leucocytes from normal peripheral human blood (< 5%). In bone marrow: No significant reaction is seen in normal bone marrow (< 5%). CD138 is absent from CD34 positive stem cells (1). In bone marrow from multiple myeloma patients, all plasma cell types are labelled, including reticular, polymorphous, asynchronous and blastic plasma cells (3).
