Beschreibung Synonyms for antigen PRAD-1, BCL-1, CCND-1 (2, 5). Summary and explantion Cyclin D1 is a 36 kDa protein (6), encoded by the CCND1 (BCL-1) gene located on chromosome 11q13 (7). Cyclins participates in the regulation of transitions through various phases of the cell cycle by associating with, and activating the cyclin-dependent kinases (CDKs). D-type cyclins bind to and activate CDK4 and CDK6, which in turn phosphorylate their downstream target, the retinoblastoma protein Rb. Expression of cylin D1 is growth factor dependent, and it appears in early G1 before cyclin E. Following its increased expression, cyclin D1 initiates cell cycle progression towards the S phase. Cyclin D may also have a CDK-independent activity as a transcriptional regulator, thus cyclin D1 is not only involved in cell cycle progression but also in cell growth (7). The (11:14) (q13, q32) translocation, an anomaly often found in mantle cell lymphoma and occasionally in other B-cell neoplasms, juxtaposes the CCND1 locus to an immunoglobulin-enhancer sequence on chromosome 14 causing overexpression of cyclin D1 (7).
Spezifität In Western blotting of recombinant human cyclin D1, the antibody labels a single band of 36 kDa corresponding to the cyclin D1 protein. No cross-reactivity with recombinant human cyclin D2 or D3 protein can be detected (5). SDS-PAGE analysis of immunoprecipitates formed between the antibody and extracts of cyclin D1-positive cell line cells shows reaction with a 36 kDa protein, corresponding to cyclin D1 (5). As demonstrated by Western blot, the antibody recognizes the cyclin D-equivalent protein in mouse (6, 8) and rat (6).

Anwendungen

Applikationshinweise Paraffin sections: The antibody can be used for labelling paraffin-embedded tissue sections fixed in formalin. Pre-treatment of tissues with heat-induced epitope retrieval is required. Optimal results are obtained with Dako Target Retrieval Solution, High pH, code S3308, or 10 mmol/L Tris buffer, 1 mmol/L EDTA, pH 9.0. The following solutions Dako Target Retrieval Solution, code S1700 and 10 mmol/L citrate buffer, pH 6.0 or pre-treatment of tissues with proteinase K were found destructive of the epitope. The tissue sections should not dry out during the treatment or during the following immunocytochemical staining procedure. Unless the stability of the diluted antibody and negative control has been established in the actual staining procedure, it is recommended to dilute these reagents immediately before use, or dilute in Dako Antibody Diluent, code S0809. Positive and negative controls should be run simultaneously with patient specimen. Frozen sections and cell preparations: The antibody can be used for labelling acetone-fixed, frozen sections, although the labelling has been reported to be weaker, and the morphology less defined than on paraffin sections (5). The antibody is also well-suited for cell preparations (5, 8). Dilution: Monoclonal Mouse Anti-Human Cyclin D1, code M7155, may be used at a dilution range of 1:20-1:40 when applied on formalin-fixed, paraffin-embedded sections of human mammary carcinoma using 20 minutes heat-induced epitope retrieval in Dako Target Retrieval Solution, High pH, code S3308, and 30 minutes incubation at room temperature with the primary antibody. Optimal conditions may vary depending on specimen and preparation method, and should be determined by each individual laboratory. The recommended negative control is Dako Mouse IgG2a, code X0943, diluted to the same mouse IgG concentration as the primary antibody. Visualization: Dako LSAB/HRP kit, code K0679, and Dako EnVision/HRP kits, codes K4004 and K4006, are recommended. For frozen sections and cell preparations, the Dako APAAP kit, code K0670, is a good alternative if endogenous peroxidase staining is a concern. Follow the procedure enclosed with the selected visualization kit. Automation: The antibody is well-suited for immunocytochemical staining using automated platforms (2). Performance characteristics: Cells labelled by the antibody display a nuclear staining pattern. Normal tissues:. The antibody heterogeneously labels normal fibroblasts (1, 5), endothelial cells (3), the epithelia of the head and neck region, epidermis, hair follicles, cervix, vagina, rectum and the transitional epithelia of the urinary tract (5). Dendritic cells have been reported to show both nuclear and cytoplasmic staining by the antibody (2). In general, normal epithelial tissues reveal low expression of cyclin D1 restricted to the proliferative zone (5).
