Beschreibung Synonyms for antigen KOC (K homology domain containing protein overexpressed in cancer), L523S, IGF2BP3 Summary and explanation IMP3 (insulin-like growth factor II mRNA-binding protein 3) is a 580 amino acid oncofetal RNA binding protein containing four K homology domains which is encoded by a 4350 bp mRNA transcript produced by the IGF2BP3 gene on chromosome 7p11.5 (1,2). This 65-70 kDa protein regulates insulin-like growth factor II (IGF-II) transcripts during embryogenesis and is later re-expressed in a proportion of neoplastic cells from various tumor types including lung squamous cell carcinoma and lung and pancreatic adenocarcinoma (3-6). IMP3 was originally identified during an analysis of differentially expressed genes comparing pancreatic cancer to normal pancreas (1). Subsequently, IMP3 was isolated using cDNA library subtraction and microarray technology as preferentially expressed in lung cancer compared to normal lung (7). IMP3 transcripts have also been detected in fine needle aspirates of patients diagnosed with colorectal and pancreatic carcinoma (8). mRNA expression was found to be minimal in many normal tissues using rtPCR and microarray analysis (5). A subset of tumors have been found to express IMP3 protein including melanoma, mesothelioma and carcinomas from the bladder, cervix, colorectum, esophagus, liver, lung, pancreas, stomach, and uterus (9-19).
Spezifität Internal studies demonstrated a 70 kDa band in Western blots of human lung carcinoma cell lysate A549 using anti-IMP3, corresponding to the expected molecular weight of the IMP3 protein (1, 5). A faint 48 kDa lower molecular weight band was also identified. Monoclonal mouse anti-IMP3 was also tested using an ELISA assay to detect immunoreactivity with the recombinant IMP3 protein.

Anwendungen

Applikationshinweise Paraffin sections: The antibody can be used for labeling paraffin-embedded tissue sections fixed in formalin. Pretreatment of tissue with proteolytic enzymes is not recommended. The deparaffinized tissue sections must be treated with heat prior to the IHC staining procedure. Heat-induced epitope retrieval (HIER) involves immersion of tissue sections in a pre-heated buffer solution and maintaining heat in a water bath (95-99 °C). Alternative heat sources m ay be used for HIER upon validation against the recommended procedure. Use a 40-minute heating protocol for HIER performed at 95-99 °C, after thermal treatment, a llow the jar with buffer and slides to cool for 20 minutes at room temperature. Rinse well with buffer or deionized water following HIER. Target Retrieval Solution pH 9 ( Dilution: Monoclonal Mouse Anti-Human IMP3, The cellular staining pattern for anti-IMP3 is predominantly cytoplasmic, with rare nuclear and membrane staining (4, 20). Normal tissues (20): Anti-IMP3 immunostains a variety of normal cell types, however the staining intensity of the majority of positive cell types is generally low. Intense immunostaining has been observed in oocytes of the ovary, glandular epithelium of the stomach and squamous epithelium and germinal center cells in the tonsil (20).
