Beschreibung Monoclonal mouse anti-D2-40 identifies a 40 kD O-linked sialoglycoprotein present on a variety of tissues including fetal testes and testicular germ cell tumors. 1 D2-40 has also been demonstrated to react with lymphatic endothelium and is unreactive with vascular endothelium. 2, 3 In neoplastic tissue, immunostaining of lymphatic endothelium by D2-40 has been shown to be useful in identifying lymphatic invasion of primary tumors. 2, 3 Specificity Introduction
Spezifität Monoclonal anti-D2-40 was tested by Western blotting of cell lysates from a variety of tumor cell lines. D2-40 identified a 40 kD band in immunoblots of HEY ovarian epithelial carcinoma cells and a slightly higher molecular weight form in blots of MGH-U3 cells. D2-40 immunoreactivity has also been observed in Western blots of SK-OV-3 and Caov-3 ovarian epithelial carcinoma cells, PA-1PA-1 ovarian teratocarcinoma cells, Tera-1 and Tera-2 testicular teratocarcinoma cells, 833 embryonal carcinoma cells, RT-4 bladder transitional carcinoma cells and U-2 OS osteosarcoma cells. No reactivity was observed in Western blots of other tumor cell lines derived from a variety of cancers. 1 The effect of enzyme digestion on the immunoreactivity of D2-40 was tested using affinity purified antigen from HEY cells and intact HEY cells. The immunoreactivity of D2-40 with Western blots of purified antigen and with cells by flow cytometry was unaffected by the removal of sialic acid residues through neuraminidase treatment of the cells or protein. Treatment of protein or cells with O-sialoglycoprotease destroyed the immunoreactivity of D2-40 in both Western blotting and flow cytometry, demonstrating that the antigen recognized by D2-40 is an O-linked glycoprotein. 1 provided

Anwendungen

Applikationshinweise This antibody is intended for laboratory use to identify qualitatively by light microscopy D2-40 expressing cells in normal and neoplastic tissues using immunohistochemical (IHC) test methods. Positive results aid in the identification of lymphatic invasion of a variety of cancers. 2, 3 Interpretation must be made within the context of the patient's clinical history and other diagnostic tests by a qualified pathologist. Refer to the General Instructions for Immunohistochemical Staining or the Detection System Instructions of IHC procedures for: (1) Principle of Procedure, (2) Materials Required, Not Supplied, (3) Storage Paraffin Sections Anti-D2-40 can be used on formalin-fixed, paraffin-embedded tissue sections. Pretreatment of tissue with proteolytic enzymes is not recommended. The deparaffinized tissue sections must be treated with heat prior to the IHC staining procedure. Heat-induced epitope retrieval (HIER) involves immersion of tissue sections in a pre-heated buffer solution and maintaining heat in a water bath or steamer (95-99°C). Use a 20-minute heating protocol for HIER performed at 95-99°C, after thermal treatment, allow the jar with buffer and slides to cool for 20 minutes at room temperature. Rinse well with buffer or deionized water following HIER. For greater adherence of tissue sections to glass slides, the use of Silanized Slides (code S3003) is recommended. Target Retrieval Solution (code S1700) or 10x Concentrate (code S1699) is recommended. Cryostat Sections And Cell Smears Anti-D2-40 can be used for labelling acetone-fixed cryostat sections or fixed cell smears. Follow the recommended procedure for the detection system selected. Staining interpretation The cellular staining pattern for anti-D2-40 is cytoplasmic and sometimes membranous.
